Though all religions are vastly different, the overall belief is geared to praise and worship God. Two religions that will be discussed in this paper that conflict with Christianity are Judaism and Islam. In Judaism, besides cultural division within, most of their conflict is with modern society. In Islam, the Muslim community has a great political outlook that has been coupled with their religious beliefs and stereotyping has been more recently a trigger for conflict. In this paper we will discuss the contemporary issues within Christianity. We will review the historical connections and theological similarities with other religions such as Judaism and Islam. We will also take a look at Christian denominations and how they interact with Buddhism. Contemporary Issues within Christianity Christians have struggles between their own religion, and with other religions beliefs. Christian’s belief in One God, God of Love. In fact for most of his life Jesus practice the Judaist faith and even celebrated Jewish holidays. Despite the fact that Christianity was a result of Judaism, Christians tended to have more freedom than Jews, while the latter was often persecuted for practicing their religion. Christianity has a connection to Israel that is almost as strong as Judaism. Nazareth which is based in Israel is considered the birthplace of Jesus, as well the birthplace of the Jewish Christian Church, however after the destruction of the church’s temple in 70C. E it seems that the church gradually faded into existence. While there may be some differences in how each religion worships and celebrates their religions. They both emphasize people being kind towards one another, believing in one God, and just being an overall good human being. The Muslim religion has points of emphasis on being peaceful, for instance; In Islam, the Muslims look to Sura 22:56 who say when speaking of religion there’s no compulsion. Though many in the world of Islam would believe that no compulsion means inside their religious world there is no competition amongst religion. Sura 29:46 says do not have a quarrel with outside religions but only if there is wrong doing. However, Sura 41:34 say to be make nice with the person doing wrong by doing good things for that person. Almost similar is the bible which has its parts of conflict with violence mostly from the Old Testament. It contests that a Canaanite society is infested with evil doings especially child sacrifice. Christian Denominations In religion, there is a power struggle to keep itself together. The ten commandments were clear on this issue. â€Å" Thou shall not worship false idols†, (Ten Commandments, 2002-2011). But there are seven billion people on this planet. Chances are that at least one of those seven billion people would want to worship another idol. From this idea, they would separate from their original church and form their own church. Worshiping whomever they found fit to be the one who deserved to be worshiped. As a matter of fact, this has happened many times in the history of Christianity and Judaism. Why are there so many Christian denominations? Well first we would have to differentiate between the non-Christian cults, and the false religions. Christian denominations are Lutherans, as well as Presbyterians. Then there are Jehovah’s witnesses and the Mormon religion, are examples of such cults. But islam and buddism are separate religions all together. Trials such as the protestant reformation gave rise within the Christian church to denominations. A movement to reform the roman catholic church that took place during the 16th century. Out of this event, four major division of traditional Protestantism emerged. These four were Lutheran, reformed, Anglican, and Anabaptist. But many more denominations would grow over the coming centuries. The denomination Lutheran was named after Martin Luther. This is based off of his teachings. Methodist was named from its founder, John Wesley. John Wesley was famous for coming up with methods for spiritual growth. Presbyterianism is named from their view on the churches leadership, and finally, Baptists. They got the name from the importants of the baptism ceremony. But not all the other denominations believe in baptism. So each one has a slightly different doctrine. Another is the availability of the lords supper to all. Or at least those who’s testimonies could be verified by the church leaders. Looking at why they have divisions, it seems clear that its not about Christ as the lord and savior. But it seems rather the differences of opinion by godly, flawed evidence, people seeking god. Seeking to retain doctrinal purity of what is their understanding of his word. Looking at today, denominations are many and vary wildly. The denominations have spawned numerous styles such as Nazarenes, Christian and Missionary Alliance, Evangelical Free, Assemblies of God, ect. * A snowmachine clutch actually consists of two separate clutches connected together by a rubber belt. o The primary clutch is connected to the engine's crankshaft o The secondary clutch is mounted on the end of the jackshaft (which connects to the drive shaft via a chain and gears). * The primary purpose of the clutch is to smoothly transmit power from the engine to the jackshaft and to remove the connection when the engine is idling so that the machine is not always rolling. * This type of system is also referred to as a continuously variable transmission. It is called this because as the engine speed increases the final drive ratio increases. That is, the difference between the engine speed and track speed decreases. It is equivalent to an automatic transmission on a car with an infinite number of gears that you never felt shift. Let's take a look at how this "infinite gearing" process works: 1. At idle the primary clutch just spins and does not "grab" onto the belt. As engine RPM (revolutions per minute) increases to engagement speed (the RPM where the snowmachine starts moving), the primary clutch begins to pull together and start squeezing the belt. 2. Now the belt is turning. This makes the secondary clutch turn, which causes the track to turn and the snowmachine to move forward. 3. As engine speed increases above engagement, the primary clutch squeezes together some more and pushes the belt so that it moves to a larger radius on the primary. My father, brother and uncle all are designers. When I was young, way before I knew what graphic design was, I would use to go to my dad’s office and be amazed by the vibe of a creative agency. At that point I knew I wanted to be in a creative field. When the time came to go to college, I had the opportunity to come to New York and att end the School of Visual Arts. During the program I got really interested in editorial design, especially book design. So much that I did my internship with Simon Schuster and my first job was designing book covers and interiors for Columbia University Press.Have you done much work with independent authors? If so, is there anything different about it compared to working with a traditional publisher?I haven’t had the chance to work with many independent authors yet, but I am excited about the prospect of working more directly with the author to create the cover. With a traditional publisher there are a lot channels you have to go through to get the cover approved internally before it even reaches the author for the first time. These three terms explain the different types of love a human being can acquire. Philia is a love of friendship, which is grounded in commonality. However, eros is a kind of love that seeks something from the other person or thing. Lastly, agape is the love that wills the good of the other and is completely self-giving. The meanings of these Greek terminologies, philia, eros, and agape, allow us to better understand and discuss our relationship with God and human fulfillment. In order to find human fulfillment, one must find God of Jesus Christ as well. Erotic loving is our desire for this fulfillment, which can be seen through eros. Human nature can be simply defined in our constant want and need for infinite. The infinite, for humans, is never being satisfied, always wanting more, the best, and as much as we can get of something. This insatiable drive within all of us takes us to the question of human fulfillment. Eros is not a noun but yet an action or an activity. For instance, eros is the longing and search for a significant other throughout our lives. By the same token, we search for God and yearn for infinite happiness and fulfillment from God. All human beings in one-way or another seek about human fulfillment. An example of someone who struggled with human fulfillment is Augustine. Although later he became known as Saint Augustine, in his earlier years he was everything but. Augustine was very intrigued by very common materialistic desires such as worldly possessions and lust. These skewed desires enthralled his thoughts, words, and actions. Consequently, Augustine became involved in petty crimes in order to feed this idea of â€Å"love† in the materialistic world. This concept of love and desire can be further illustrated as a receding horizon; the farther you continue into the horizon the bigger and more numerous your wants become. Later on Augustine reflected upon his materialistic ways of life and made significant life changes to become the Saint Augustine he is referred to today. As exemplified, the essence of eros can have a negative outlook and has been evaluated as such by Jean Paul Sartre. Sartre attested that the human being is a useless passion due to our insatiable desires. Markedly, human beings desire to retain the infinite and that need is simply impossible. For the search for the unlimited is tiresome and unappeasable. Conversely, there is an alternate way of looking at eros. In order to have hope, God has to appear to human life as not a receding horizon but an absolute savior. Human beings long for the truth and due to the constraints of science and history, Jesus is seen in an alternate way because he is no longer limited by space or time. In this case, Karl Rahner and Saint Augustine both agree that God always initiates the relationship with human life. Namely, they asserted that we are constantly drawn to God and God is not drawn to us. Because of this, we have the option of forming a better relationship with God. Rahner said that God dispenses his grace to all human beings of every religion in and God is continually accepting. Chiefly, it is not God’s choice to accept or deny us, for that is not his matter?. Anthony De Mello describes another explanation of human fulfillment to us. He explains to us that in order to have human fulfillment, we must detach ourselves from our worldly attachments. As human beings, we allow ourselves to believe that we cannot be happy without a certain thing or person in our life. Thus, that is not correct. We completely wreck ourselves with the belief in finite things. De Mello also emphasizes that we shouldn’t not care about these things and people in our life but rather we realize that we can live without them and be intent with that. ? Besides eros being a description of love there is also philia. Philia is the love of friendship or brotherly love. This kind of love is seen most commonly between friends who share things in common. Once we seek God, we have a friendship with God. Although, God is not described by philia because it is not completely reliable due to changes in friendships in positive and negative ways, and God’s love never changes. On the contrary to philia, agapic love is God. The terminology of agape explains God’s love as it wills the good of the other. Agape is all self-giving and never asks for anything in return. When a person gives them self to God, this love is agape. Nevertheless, agape and philia types of are critical to having a strong relationship with God. ?  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚   The doctrine of the Trinity can be viewed as God invites human beings into the inner life of God. Consequently, the doctrine of the Trinity correlates with this topic of discussing the types of love. The idea that God is three persons, yet one essence exhibits that God revealing Godself. God not only communicates with us, but he also draws us into the Trinity. Rahner expressed that the doctrine of the Trinity â€Å"is not a subtle theological and speculative game but is about the self-communication of God. † In the final analysis, there are two models that Christian theologians have formulated in an attempt to address the question of the salvation of non-Christians. These models are the Replacement Model and the Fulfillment Model. The Replacement Model mainly represents the Fundamentalist and Evangelist churches which existed over most of Christian history. This model emphasizes the scriptures from a literal standpoint. According to the Replacement Model, in order to be saved one must be in connection with the Church. Quite clearly, the Replacement model is not very accepting of other religions and believes only Christians will have salvation. ? The Fulfillment Model represents the majority of modern day Christians. The Fulfillment Model preaches the perspective that Christians need to have dialogue with non-Christians. In addition, many Christians think it’s not fair to punish those who have never learned the religion of Christianity or even heard of it. The Fulfillment Model is a way to accommodate people of all different faiths and as long as they are moral, that they too will see eternal life with God. By and large, Karl Rahner’s ideas mostly correlate with the ideas of the Fulfillment Model. The only real difference is that he states is that other religious views are also true. The church did not accept Rahner’s view because quite simply the church thought that other religions couldn’t be true and by believing so it devalued the image of Jesus in the Christian Church. In Vatican II, the council agreed with Rahner’s except they said there was no such thing as â€Å"anonymous Christians† and that other religions cannot be viewed as ways of salvation. Moreover, I believe that the Fulfillment model is the most accurate description of how non-Christians should be viewed. I believe that all people have salvation in God because God knows and loves all human beings. Lastly, I believe that by thinking that God doesn’t save non-Christians limits the idea God’s everlasting love.

The DSM is conceptualized as a legal system where the same rules apply to countries on an equal basis, and where cases are judged according to their merits. This principle is incarnated in the AB, which has developed a case law with this emphasis.  A final set of observations from this literature focuses on biases and inequalities within and between institutions managing trade, including the WTO in general and the DSM in particular (Busch and Reinhardt, 2003; Shaffer, 2003). Here, the main problem identified is that the DSM (and the WTO) has become too technically complex and demanding for most developing countries to use effectively in the absence of adequate assistance. Originally, this is the observation that there are too much law and too little politics in the system.   The implicit biases of systems of trade rules, including the DSM, in favor of powerful countries - reinforced through the dominance of judicial forms of rule-making - have been noted by some contributors to the literature (for example, Busch and Reinhardt, 2001).  Initial studies of developing country participation in WTO dispute settlement found that they were unable to use the system effectively to settle cases, although they had expected levels of success when cases went before panels and the Appellate Body.161 This suggested that there was a need for more training and assistance to developing countries to enable them to handle the initial stages of dispute settlement more effectively. More recent experience suggests that at least the major developing countries - and quite a few Latin American countries in general - have become more effective in using dispute settlement overall, including in using it to obtain early settlements. The establishment of the Advisory Centre on WTO L aw and the dispute settlement training activities of the WTO Secretariat and others should help ensure that developing countries continue to made progress on this front (Busch and Reinhardt, 2001).

Effect of Oxidative Stress on HAGE Expression Cancer is a group of diseases which involves uncontrolled cell growth, metastasis and invasion of cells to other tissues via lymph node or blood. Cancer is caused by many factors such as (i) Chemicals such as tobacco, alcohol, asbestos etc., (ii) Viral infections such as Human Papiloma Virus, Epstin Barr Virus, hepatitis B, C etc., (iii) Ionizing radiations such as X-Rays, UV-Rays, Gamma Rays etc., (iv) Hereditary factors such as mutations in BRCA 1 and BRCA 2, p53 mutations, APC gene mutations, Retinoblastoma gene mutation etc., (v) Hormones which promote growth proliferation such as growth hormones, estrogen, progesterone etc. Cancer is classified into malignant and benign tumours. Malignant tumours are mostly liquid tumours which metastasize to other tissues via lymph node and blood. Benign tumours are mostly solid tumours which stay at one particular tissue and seldom migrate. Malignant and Benign tumours are re classified on the type of cell and tissue from which they originate. They are classified as follows: i) Carcinomas: These are the malignancies of the epithelial cells, which line the internal organs of the body and also present on the surface the body such as skin. ii) Sarcomas: These are solid tumours present in connective tissues such as bones and lymph systems (Crowley 2006) Furthermore the tumours are classified according to (i) site of origin as in lung cancer, breast cancer etc., (ii) type of cell such as rhabdomyosarcoma and acute lymphocytic leukaemia. (Ruddon 2007) The progression from a normal cell to malignant one involves the dysregulation of genes implicated in the control of normal proliferation / death over many years. These are normal functions regulated by proto-oncogenes and tumour suppressor genes. Proto-Oncogene And Tumour Suppressor Gene Protooncogene is found to be normally expressed in all vertebrates including humans. It is also found to be expressed in some insects and yeasts. Protooncogenes play an important role regulating normal cell growth and differentiation. They also perform functions such as signal transduction and mitogenic signal execution RAS, MYC, TRK, ERK are some examples of protooncogenes (Novakofski 1991). Oncogenes are derived from protooncogenes during the process of carcinogenesis. The formation of active oncogene leads to abnormal cell proliferation and thus contributes to the formation of tumour (Alitalo and Schwab 1986). A protooncogene can be converted into an oncogene in a variety of ways such as (i)Transduction induced by retroviruses which cause the integration viral DNA with the host. This DNA on translation give rise to viral proteins which cause the activation of protooncogene, thereby leading to cancer. (ii) Mutation within the protooncogene leading to an increased activity of the pr otein. (iii) chromosomal translocation where the gene for proto oncogene is translocated to other loci, which cause its abnormal expression. For eg in the translocation of abl in chromosome 9 to bcr region chromosome 22 as found in Philadelphia chromosome (Rowley 1973) (iv) Amplification induced by gene duplication, mis-regulation of gene which leads to overexpression of protooncogenes. (Todd and Munger 1999) Unlike oncogenes, the tumour suppressor genes normally inhibit cell growth in variety of ways such as (i) repressing the genes important for cell cycle progression. For eg., pRb (retinoblastoma ) controls the G1 stage by being hypophosphoryated and not allowing the release of transcription factor E2F which promotes G1-S transition. (ii) Stopping cell cycle on detecting DNA damage. DNA damage induces p53 which now leaves from its interacting partner Mdm2 and increases the activity of p21. This p21 protein then inactivates cyclin dependent kinases which are essential for cell cycle progression, (iii) Apoptosis may also be promoted by p53 when damage is irreversible. (iv) inducing DNA repair proteins to repair DNA damage and prevent abnormal cell proliferation. (Yoshida, et al. 2000) There are a number of mechanisms by which a cancer cell survives in a body. This is illustrated in the diagram below.(Weinberg and Hanahan, 2000) Treatment The choice of therapy depends upon the location and stage / grade of the tumour, as well as the health of the patient. There are a number of therapies for treating cancer such as surgery, chemotherapy, radiaotherapy, hormonal therapy etc., The deregulation of many genes in cancer cells leads to the over-expression of altered proteins which can be used as biomarker which can provide help for the diagnosis and / prognosis of the treatment but can also be used as potential target for immunotherapy. Cancer antigens and their classification Tumour antigen is an antigenic substance produced in the tumour cells and triggers an immune reaction in the host. They are useful in identifying tumour cells and are used in cancer therapy. Tumour antigens are classified as Cancer testis antigen, Differentiation antigens, Tumour specific unique antigens, Overexpressed Self Antigens, Viral antigens, Post-translationally, Oncofoetal antigens, Idiotypic Antigens as shown in the table below Copied from (Li, et al. 2005) Amongst all these categories, cancer testis antigen represent the most promising group of antigen to be used in future immunotherapeutic interventions due to their restricted expression to mainly to tumour cells with the exception of placenta and testis which are immune-privileged sites and therefore would not pose any real issue for the risk of autoimmunity.(Simpson, et al. 2005) HAGE HAGE also known as DDX43 and CT13 antigen belongs to this category of antigens and was first identified by Martelange et al using cDNA subtraction approach of a human sarcoma cell line.(Martelange, et al. 2000)Using rational hybrid analysis, it was found that HAGE gene is located on chromosome 6. HAGE was found to be overexpressed in several tumours. There is a low expression of HAGE in normal tissues since the expression is regulated by hypermethylation HAGE is also over expressed in tissues such as testis, placenta, and ovaries. HAGE is usually expressed at the m-RNA level though recent studies in melanoma cells suggest that HAGE could also be expressed at the protein level.(Mathieu, et al. 2010) The name DDX43 comes from the fact that HAGE also belongs to another group of protein known as Dead box proteins and referred to as DDX43. (Abdelhaleem 2004) Dead box proteins These proteins were discovered in the late 1980s. (Gorbalenya, et al. 1989)Dead box proteins belong to the family of RNA helicases and play an important role in the transcription, post transcriptional modifications such as splicing, transport, translation, decay and biogenesis of ribosomes. NTP hydrolysis provides energy to these enzymes and they unwind dsRNA or disrupt RNA-Protein interaction. DDX and DHX are widely studied Dead Box Proteins. There are many classes of DDX and HAGE belongs to DDX43. They are so named because they contain the amino acids D-E-A-D (Asp-Glu-Ala-Asp) in one of their motifs namely Motif II. Besides they also contain motifs such as motif I, Q-motif, motif VI, motif Ia, Ib, III, IV and V as shown in the diagram below. These motifs bestow properties such as ATP binding and hydrolysis, RNA interaction, remodelling activity etc to these proteins. In normal tissues HAGE expression is regulated by DNA hypermethylation, Histone modifications such as histone aceylation, histone deacylation and histone methylation. However in cancer, there could be many mechanisms of upregulation of HAGE. The most commonly thought mechanism is the demethylation of DNA which leads to HAGE over-expression. (Roman-Gomez, et al. 2007) HAGE could also be induced due to transformation of normal cells during cancer or due to the action of oncogenes. HAGE might also be induced randomly. HAGE may play a role in pre-mRNA splicing, ribosome biogenesis, transcription and initiation of translation. (Rocak and Linder 2004) This project will focus on Head and Neck Cancer. Head and Neck Cancer Head and Neck cancer are the cancers that arise from the upper aerodigestive tract such as nasal cavity, oral cavity, pharynx, larynx and paranasal sinuses. Most of them are of squamous cell carcinoma type of the head and neck and have been given the acronym -SCCHN or HNSCC. (Argiris, et al. 2008). The diagram of the anatomy of the head and neck is illustrated below Copied from (Wu, et al. 2009) More than half a million patients are diagnosed with head and neck cancer every year. Head and Neck cancer in fact accounts for more than 3-10% of the cancers(Gourin and McMains 2005). Smoking, alcohol, chewing of betel leaf with tobacco and areca are the major risk factors for this cancer. Recently, HPV type 16 has been identified as one of the causes for head and neck cancer and account for 40% (Goodger and McGurk 2000) The progression of this tumour is complicated. It progresses from the normal features to hyperplasia, mild dysplasia, moderate dysplasia, severe dysplasia, carcinoma, invasive carcinoma and metastasis. The progression may be caused by due to genetic instabilities such as loss of heterozygosity of chromosome 9p21, inactivation of of p16 and loss of 3p, loss of 18q, inactivation of PTEN, some translocations, 17Beta heterozygosity or TP53 mutation. (Argiris, et al. 2008) Copied from (Argiris, et al. 2008) Interestingly HAGE was found the John van Geest group to be overexpressed in 40% of HNSCC cancers (unpublished data) and although much of its function is now emerging no information exist at the moment regarding the regulation of its expression. However it has been observed that cells left for longer period of time in the incubator had a significantly higher expression of HAGE at the mRNA and protein level. Aim of this project The observation that HAGE was over-expressed in 40% of HNSCC led to hypothesis that cells under stress i.e lack of nutrients, decrease oxygen level, as well as other factors influence HAGE expression. Moreover it has been found that HAGE is also expressed in 20% of Acute Myeloid Leukemia and 50% Chronic Myeloid Leukemia as well as many solid tumours such as HNSCC (Adams, et al. 2002). This suggests that there must be something in common between these 2 very different forms of cancer. CML and solid tumours are known to have a high expression of reactive oxygen species (ROS), therefore it might be possible that ROS induces HAGE expression. Thus this project will focus on the effect of oxidative stress has on HAGE expression. The effect of other stress such as temperature may also be investigated. METHODS Cell Culture The PCI 13 and PCI 30 cell lines would be supplied by Prof E Tatour. These cell lines are specific for Head and Neck cancer. The cells would be grown in RPMI 1640 +10%(v/v) FCS+2mM L-glutamine and incubated at 37oC and 5%C02 atmosphere. Extraction of m-RNA The culture media would be removed and washed with DPBS(Dulbeccos phosphate buffer saline). Then DPBS will be removed followed by the addition of RNA-STAT 60. RNA pellets would be then retrieved and re-suspended in ddH2O. RNA extracted is then quantified using NANODROP 8000 UV spectrophotometer. RNA would be then added in measurement wells of the spectrophotometer. The spectrophotometer would be read at 260 and 280nm and the amount of RNA will be calculated. The RNA concentration is calculated in  µg/ µl before being adjusted to 1 µg/ µl by ddH2O. RNA samples would be then stored at -80oC and used for conducting RT-PCR and RTq-PCR in the future. RT-PCR (Reverse Transcriptase PCR) From the extracted RNA cDNA will be synthesized. 2  µg of RNA sample will be mixed with 1  µl of oligo-dT primers and diluted to 15  µl of ddH20 in an Eppendorf tube. All RNA samples would follow the same process. UNO Thermoblock will be used to heat the tubes to 70 °C for 5 min which allows the primer-RNA annealing. 10  µl of reverse transcriptase mix, which will be prepared by mixing Muloney Murine Leukemia Virus 5-X buffer, dNTPs, RNasin RNAse inhibitor, M-MLV-reverse transcriptase and ddH20. The tubes will be then pre-heated in a water bath at 39.2 °C for 80 min which allows cDNA synthesis. Tubes will be removed and kept again in the UNO-Thermoblock and heated to 95 °C for five minutes which stops the reaction. These tubes would be stored at -20 °C for future use. Real Time qPCR cDNA generated from RT PCR will be used as a template and the reagents used will be (i) 6.25 µl iQ Sybr Green, (ii) 0.5 µl gene specific sense primer (iii) 0.5 µl gene specific anti-sense primer, (iv) 4.75  µl ddH20 and 0.5 µl cDNA template. These reagent mixtures will be added to all tubes for a specific gene. Samples will be usually carried out in duplicate with a negative control that contains the reaction mixture without cDNA. Rotogene 6000 real-time qPCR analyser will be used to carry out RT qPCR. Relative gene expression using 2ΔCT method will be calculated with the help of the expression of housekeeping genes HPRT-1, HSP-27 and HSP-90. The primers which will be used in real time qPCR experiment is given below. These primers are supplied by mwg-Eurofins. The primers have a stock concentration of 100pmol/ µl and will be diluted to a working concentration of 10pmol/ µl. From this working 0.5 µl (5pM) of primers will be used. Total Protein extraction Cells will be grown to 75% confluence in T75 flasks. Cells will be trypsinised, washed and re-suspended in DPBS and these cells will be then counted using trypan blue. Cells will be diluted to 1X106/ml using DPBS and 5 X106 cells would be taken in a 1.5ml eppendorf. Centrifugation will be done to pellet the cells. The supernatant will be discarded and lysis buffer containing a cocktail of RIPA buffer and 10% protease inhibitor will be added to the pellet. The eppendorf tube will be then placed on a tube rotator at 4 °C for 30 min. It will be then kept on ice for 30 min. Cells will be again centrifuged at 14000 RPM for 30 min at 4 °C. Aliquot of the supernatant will be prepared to be stored at -20 °C for future use. Total Protein Assay Total protein assay will be carried out using BioRad Dc protein assay reagents. Series of dilutions (0.2,0.4,0.5,0.8,1.0,1.5,2.0mg/ml) will be created using stock BSA solution with the concentration of 10 mg/ml. Protein extracts and standards would be tested in triplicates and duplicates respectively. Assays will be performed in 96-well round bottom plates. 25 µl and 200  µl of reagent A and B will be added to all the samples and the samples will be incubated at room temperature for an hour. Proteins will be then read at 750nm and their concentrations would be calculated based on the standard values. Western Blotting 30  µl of sample will be loaded into the wells of SDS PAGE gels. 1X tris-glycine-SDS will be used as the running buffer. A known molecular weight ladder will also be run alongside the samples. Initially 70V current will be applied to aid the migration of proteins through the 5% stacking gel. Once the proteins reach the 10% separating gel, 90V current will be applied. After the proteins run through the gel, they will be transferred to the PVDF membrane with the help of liquid transfer. This membrane will be prepared by washing with 10% methanol for 5 seconds, ddH2O for 5 min and transfer buffer for 10 min. Liquid transfer will be carried out using the following steps. 1. Cold transfer buffer will be used to immerse a gel frame. To this gel frame a sponge pre soaked in transfer buffer will be placed Then a filter card, gel, PVDF membrane, another filter card and a second pre soaked buffer sponge will be placed in order starting from the first filter card. Proteins will be transferred with the help of electric current of 100V applied for one hour. After completion, membranes will be cut and treated with different antibodies.10% (w/v) Marvel milk solution will be used to wash the membrane for one hour at room temperature. This washing blocks the non-specific binding sites. The blocking solution will be then discarded and the antibody diluted to 10% (w/v) in Marvel milk solution will be added. This antibody coated membrane will be agitated overnight on a plate rocker at 4 °C. The next day there would be 3, 10 min washes with TSBT(Tris-Buffered Saline-Tween-20). During these washes the membrane vessel will be agitated in plate orbital shaker at room temperature. After completing the washing process, a secondary antibody specific to the primary antibody will be added. The steps for adding the secondary antibody are same as the primary antibody addition except for the fact that no overnight incubation is required and the secondary antibody will be incubated only for one hour while being agitated on the shaker at room temperature. The marker will also be stained using streptavidin-HRP secondary antibody. After performing the above step the membranes will be developed. Membrane development will be performed by placing the membrane in a tray and washing it with ECL reagent. The membrane would be then exposed for a certain period of time using CCD camera. Assay for ROS-DCFDA Stress Test Stock Solution Preparation The stock solution of Hanks Buffered Salt Solution (HBSS)will be prepared according to the manufacturers protocol. DCFDA(2,7-dchlorofluoroscein -diacetate) Assay To test ROS levels DCFDA test will be performed. Before conducting the DCFDA assay, the standardization of optimum concentrations of H2O2 and DCFDA for detecting cell stress should be performed. It is important to note that H2O2 should be added only sub-lethally and should not be added in proportion which may cause cell death. The PCI 13 and 30 cells will be plated out in two 24 well plates. 1ml of each type of cells plus RPM1640 10% FCS media will be added to 8 wells per plate and will be incubated overnight at 37 °C. The media will be removed the next day and increasing concentration of DCFDA will be added to both plates and the cells will be incubated with Tinfoil wrapping to prevent light exposure and incubated at 37oC for 30 min. The cells will be then washed and then cold HBSS will be added. After this step the cells will be stressed with increasing concentration of H2O2 (see diagram below). The cells will then be again wrapped in tin foil and placed on a rocking platform for 15 min. The same method will be followed for treating other wells with different concentrations of DCFDA. Experiments will be performed at least twice for each cell line. H2O2 will be removed after 15 min and cells will be trypsinised with 100microL trypsin and Versene. 800 microL of DPBS will be added to one of the 2 wells an d will be pipetted thoroughly so as to remove the cells from the well surface. This will be then transferred to the other well and then 1ml of solution would be transferred to Flow associated cell cytometry (FACS) tube for analysis. The same procedure will be carried for other sets of wells. DCFDA fluorescence would be measured using Gallios flow cytometer and results would be analysed in Kaluza program. Time Line For The Project During the month of May, all techniques such as Western Blotting, mRNA/protein extraction, Real Time PCR will be learnt. This time is indicated in red colour in the graph below. This will overlap with the time duration, from May to the end of June, during which hydrogen peroxide experiment will be performed as per written in the method section and the expression of HAGE will be monitored at both the mRNA and protein level. Also from the start of June to the end of July repeat experiments will be performed and if time permits, the effect of thermal stress on HAGE expression will also be investigated. This is indicated in the graph below. SUMMARY Cancer Testis Antigens (CTA) are antigens that are expressed in a variety of tumours. They are usually absent in normal tissues with the exception of testis and placenta where they are expressed as self tolerant antigens. Since these antigens have strong immunogenicity and their expression is mostly restricted to tumours, they are ideal targets for cancer immunotherapy. Therefore much research is ongoing for the identification of CTA. Recently helicase antigen HAGE was identified as a CTA and was found to be over-expressed in HSNSCC, AML and CML. Thus there might be a common link between these different types of cancers. Moreover ROS is known to be induced in all these tumours and it may be possible that ROS causes increased expression of HAGE. Thus the aim of this project would be to investigate the effect of oxidative stress on HAGE expression. HAGE expression would be analysed both at the m-RNA and protein level in two cell lines namely PCI 13 and PCI 30 using techniques such as m-RNA extraction, RT PCR, Western Blotting and Protein assay. The ROS level would be evaluated using the DCFDA assay. Since the mechanism of HAGE regulation is unknown, positive result in the project would help in elucidating a mechanism by which HAGE could be regulated.

This quote concludes the beautifully written ballad located in the first chapter of Margaret Atwood’s Alias Grace. By summarizing the events leading up to the murders, the murders themselves, and the ensuing trial, the poem presents the reader with what appears to be a foreshadowing of things to come. However, though the ballad reflects many of the novel’s events, there are several differences which contradict Grace’s narration.   Ã‚  Ã‚  Ã‚  Ã‚  The poetic verse and the story told by Grace contain numerous similarities. As the ballad states at the beginning, Grace says she was sixteen years old when the murders at the house of Thomas Kinnear occurred; James McDermott worked as a stable hand, and Grace was the serving maid. Also alike is the poem’s description of Nancy as a â€Å"no well-born lady†¦.who goes in satin and silk, The finest ever seen† (11). When first meeting Nancy, Grace wonders why â€Å"a housekeeper would be wanting a dress like that,† (200) immediately noticing Nancy is dressed rather well considering her occupation. When the murders take place in the novel, James strikes Nancy on the head with an axe and throws her into the cellar where she eventually died with an unborn baby in her womb. This event was depicted in the poem, as was the scene where James and Grace steal valuables from Mr. Kinnear’s house and fled across the lake to the Lewiston Hotel in t he United States. As the ballad progresses, the two are later arrested at which point Grace states she does not remember seeing the murders take place. Also similar, is James’ declaration of Grace being the one who lead him on, and if not for her the murders would have never happened. When the poem explains how Jamie Walsh marked Grace a murderer at the trial, yet she was given a life sentence while James was hung and dissected at the University, Grace’s tale is reflected perfectly. The ballad concludes with Grace receiving forgiveness and entering a life of paradise. This appears apparent at the end of the novel as Grace is pardoned, and then fulfills her â€Å"apple skin prophecy† of marrying a man with a first name beginning with ‘J.’ Though the above events are comparable to the story Grace tells, the ballad contains several discrepancies as well.

This can have dramatic negative effect on teamwork and one can see a how a metaphor can be drawn between this and troops that go to war do not want to make friends with someone who may die the next day. 1. Studying English language in an English-speaking country is the best but not the only way to learn the language. Do you agree or disagree with this statement. Some people believe that the students have to go abroad to study English while other people claim that beside learning English in an English-speaking country, there are a number of ways to study this language. I am strongly persuaded by the latter idea by the following reasons. First of all, it cannot be denied that learning English in an English-speaking country have many advantages.While overseas, students will have opportunities to practice listening and speaking with British people, which is conducive to the development in using language. Moreover, by dint of living in foreign countries, students can also experience the culture first-hand (have first-hand experience in foreign culture), which is a great help when trying to understand the language. For example, living with the native family will provide the students oppor tunities to discover about new cultures and customs. Besides this, if student attend language full time, teachers will be native speakers.In that case, not only will student’ speaking and listening skills improve but the attention can be given to develop reading and writing skills. However, the fact has shown that, most students in non-English-speaking countries often study English at secondary schools, sometimes at universities. Although the spoken English is not usually of a very high standard, the knowledge in grammar is often quite advanced, which will be good basics to perfect language ability in the future. It is obvious that the more technical science upgraded the more ways students can approach the knowledge worldwide without going abroad.Equally important, learning English basics at secondary school is much less stressful than studying language while overseas. I am the Man in the â€Å"Love Poem† I consider myself to be the narrator in John Frederick Nims’ Love Poem because it immensely reflects my current feelings for a particular person. Nims refers to his lover as his â€Å"clumsiest dear,† (Nims, 2003, p. 67) which quite bears a resemblance on the nature of my own special someone. His subject’s palms are compared to the â€Å"bulls in china, burs in linen, / And have no cunning with any soft thing† (Nims, 2003, p. 67). This metaphor that insinuates roughness and clumsiness also reflects some of the characteristics of my lover. Being a â€Å"Misfit in any space. And never on time. † (Nims, 2003, p. 67) surely creates an image of a person entirely different than other people. He or she does not always fit in any common group of people and is expected to break most rules. However, like the narrator’s last two lines in the fourth stanza, â€Å"In traffic of wit expertly manoeuvre / And keep us, all devotion, at your knees. † (Nims, 2003, p. 67), I still find myself adoring him for his remarkable wisdom. Simply put, despite all the imperfections of this particular person, I still love him or her without any doubt. However, it is also important to consider that the relationship in this poem does not only include the woman and the blonde but also Jack whom the woman has been with for a long time. This is validated on the 3rd and 4th line of the second stanza where the narrator introduces him as, â€Å"the other, jack, had a claim / On her already . . . / He understood her. His long, lithe frame† (Farley, 1937. n. p. ). Hence, it is clear that there are two stages of love in this poem in dependence to whose relationship in the love triangle we are speaking about. Two Figurative Languages that Compares the Love in â€Å"The Lover Not Taken† The first figurative language is a metaphor that speaks of the new guy’s physical description as â€Å"smooth as a yellow wood† (Farley, 1937. n. p. ). His physical beauty is compared to that of the yellow wood which can mean several things like the end of summer or the beginning of fall which can also be considered to be nice to look at. Perhaps, it can be a literal comparison of the man’s smoothness to a smooth yellow tree. The second comparison is on the first line of the second stanza. The narrator’s statement â€Å"She liked his hair,† (Farley, 1937. n. p. ) obviously compares the superficial relationship of the woman and the blonde. If one would deeply analyze, the attraction of the woman to the blonde is too superficial or simply physical. Liking his hair and smile obviously just symbolizes her extreme lust for the blonde guy that she is actually willing to risk her long-term relationship with a complete stranger who has a beautiful smile and hair. The Lover Not Taken is Most Likely Doomed to Misery As for my opinion, guilt leads a person to misery. In this poem’s case, it is most likely for the woman’s relationship with Jack to be miserable if guilt would consume the woman’s relationship with him. Honesty and loyalty to a partner are two of the most important elements that determine the stability of a relationship. However, since this poem consists of two relationships of a particular woman; the answer would still depend on whom she would end up with. If she ends her relationship with Jack and start a new one with the blonde, there is a possible occurrence of a â€Å"happily ever after story†. On the other hand, if she stays with Jack and continues her affair with the blonde guy, it is most likely to end up in misery. The Tone of â€Å"The Lover Not Taken† By analyzing the atmosphere being suggested on the first stanza of the poem, â€Å"And, mulling it over, long she stood, / Alone on the road, loath / To leave, wanting to hide in the undergrowth. † (Farley, 1937. n. p. ), the initial seriousness of the narrator’s tone is evident. There is too much concentration on the woman’s part as she mulls over her future decision whether to have an affair with another man or not. However, the tone changed abruptly on the last two lines where she suddenly stopped contemplating on which decision to make by taking the fast way home and phoning the blonde. Setting of â€Å"The Lover Not Taken† Obviously, this poem is inspired by Robert Frost’s famous poem â€Å"The Road Not Taken† which tackles the difficulty of deciding what path to choose in life. However, Farley similarly illustrates the difficulty of making decisions by pointing out a particular situation such as being torn between two lovers. Like Frost’s original piece, a person is facing a certain road pondering about a situation he or she needs to handle. By basing the setting and theme on Frost’s poem, Farley has further emphasized the central message of Frost’s poem. Since, it is inspired by Frost’s â€Å"Road Not Taken†, it is important to consider not only why it is set in that particular setting but more importantly why Farley chose Frost’s poem to communicate her own message.
